Many characters can be rescued from the Gungeon or merely reside in the Breach. Some will appear at both locations. The following is the run-down of who they are, what they do, and how it helps you to know them!
Ser Manuel
The knight who watches over the Halls of Knowledge. He will teach you how to survive in the Gungeon and is also your first challenge. He isn't a threat to you, especially since he is already dead after being shot in the back by Blockner. He is only found in the Halls of Knowledge until you discover his secret and learn about Blockner's betrayal. Defeating Blockner will cause Ser Manuel to appear in the Breach where he'll reward you with the Betrayer's Shield and the Badge.

Shopkeeper
The owner of the stores in most of the chambers. He sells you items for a fair price, but if you fire a weapon too many times in his store or get caught stealing then he'll open fire, take all the items on sale away, and kick you out.
Resourceful Rat
This sneaky rodent will take anything you leave behind. If you leave ammo, armor, items, or guns laying on the floor of a room that you are no longer in then the Resourceful Rat will steal it and leave a note thanking you for being a chump. If you come into a room while he is stealing you can shoot him to interrupt his thievery.
When taking a shortcut via the Tinker to the Black Powder Mine, Hollow, or Forgeyou'll see him in a disguise to offer you a free gun.
Drunkard
This blue creature sits outside the Halls of Knowledge and tells you tales about his companions in the Breach as well as the bosses you've defeated.
The Tinker
The engineer who fixes the elevators in the Gungeon. He'll be found in the Gungeon Proper entrance waiting to meet you before you can speak to him in the Breach. Once you have a shortcut established to the second Chamber he'll open a room in the Breach for you to access these shortcuts.
Cadence & Ox
The duo can be rescued in the Gungeon which will allow them to open a store in the Breach where you can purchase new guns and items with Hegemony Credits to unlock allowing them to appear in the Gungeon. The robotic golem, Ox, will sell you the goods while it's mistress sits in the back kicking back.
When you buy out a majority of the store the inventory will be replenished next time you go to the Breach. The following are the purchases that can be made.

Ox's arm
When you meet Ox you'll notice that one of his arms are missing. You'll be able to find his arm downin the Forge with the Blacksmith. When you find it, a balloon will pick it up and float it out of the room. It'll actually end up in the Hollow and you'll need to find it during a future playthrough. You'll need to keep finding it and sending it up one floor until it finally ends up in the Breach.
Professor Goopton
This salesman speaks a strange language that cannot be understood, but you should rescue him anyway. Doing so will bring him to the Breach and also makes him randomly appear in the Gungeon to sell you strange guns and items that usually involve fluids (water, gas, poison). In the Breach you'll be able to purchase guns and items to unlock permanently:

Winchester
This strange man can be found in the Gungeon running a shooting range game where you'll be tasked to shoot displays of his head to win a chest. The better you perform in the game will lead to a better chest. He'll never give you a Mimic Chest, but if you hit none of his targets then you'll get nothing. Meeting him once will be enough to have him appear in the Breach, but he won't talk to you until you defeat the High Dragun.
Blacksmith
The Blacksmith is only found in the Forge, the fifth Chamber of the Gungeon. She operates similarly to the Shopkeeper, but a number of quests can only be fulfilled with her help.
To properly kill The Past, you'll need the Bullet to Kill the Past. She will assemble it for you once you give her all the parts for it over the course of multiple playthroughs. Once you have given her all the parts you need to talk to her each time you want to kill The Past of your character to get the bullet needed. Otherwise, your run will end after beating the High Dragun.
You can also unlock The Robot by bringing the Busted Television to her. Lastly, the Blacksmith has a new arm for Ox in her shop.

Sell Creep
This slimy creature appears sometimes in the normal shops of the Gungeon. If you drop a gun or item on his grate then you'll get coins in return. Your cannot give him your starting gun with infinite ammo, but everything else (including Junk) can be sold.
Ledge Goblin
This NPC will sit on the right side of Breach after being rescued from the Gungeon Proper. If you walk up to the helmet next to the Ledge Goblin you can kick it off the ledge. The goblin will go down into the Gungeon to retreive it and you'll need to rescue them again. Repeating this will make the goblin go deeper into the Gungeon each time. Eventually, the Ledge Goblin will give you the helmet after having to retrieve it a number of times, adding it to your collection of items.
The Lost Adventurer
This hero with a map can be found in the Gungeon after being rescued. After saving him you might see him around the five different chambers. He'll ask you to explore the entire chamber for him wherever you meet him so that he can properly chart out the floor on his map. Doing this will give you a reward in the form of a gun or item to use. If you help him map out every chamber then he'll meet you up in the Breach.
Gunsling King & Manservant
This regal fellow and his less handsome companion can be rescued from the Gungeon and then found in the Breach to talk to. More importantly, once rescued they can be found in the Gungeon to offer you a challenge. They will give you a condition to meet for the next room when facing enemies and have you wager money on it. If you succeed in completing their challenge then you'll get your money back as well as a gun or item to use in your run.
Frifle & the Grey Mauser
Cursula
This mysterious woman, once rescued, will sell you plenty of wonderful items and guns that can prove very helpful. However, Cursula is not exactly trustworthy as all items and guns she sells to you are cursed and will increase how cursed you are.
Flynt
This living lock can be rescued from the Gungeon Proper. Once he is saved he will appear in the Gungeon at random to sell you items related to chests, but he will not accept your coins. You may only purchase his items with keys.
Trorc
Old Red
This old bullet can be rescued from the Gungeon, but his eyesight is failing him in his old age. He believes you too are a bullet and agrees to sell you things if you can find him again in the Gungeon. All the things he sells you are usually related to blanks.
Patches & Mendy
These ladies can be saved in the Gungeon. This makes them available to help you if you can find them later on in the Gungeon by practicing medicine on you. They were kicked out of their medical school for their unusal practicies, but they get the job done. Talking to them will make them smash a jar over your head and out comes a fairy that heals you completely.
Sorceress
